TGGE or temperature gradient gel electrophoresis is a useful technique for mutation detection. PCR fragments differing in a single position only can be separated quickly by electrophoresis on a linear temperature gradient. The TGGE system relies on the melting characteristics of biomolecules to separate the samples. TGGE reliably detects DNA mutations in a 1:10 dilution (and higher) of wildtype DNA making it a powerful tool in mutation screening applications. |
